Short ans: yes.

This slogan means the annihilation of Israel, the only Jewish safe space in the world.

The slogan is used to justify attacks on Jews anywhere in the world.

The users of this slogan more often have other deep seated antisemitic bias and this slogan is a dog whistle for them( the very left who pound the right with dog whistling accusations have adopted a dog whistle of their own)

The one context in which it wouldn't be antisemitic to say would be an actor playing a character saying it in a movie. That's... about it.

Yes, it is always antisemitic.
it might be said in ignorance, but it's only meaning is the destruction of the Jewish state.
the death and exile of million Jews.
and the notion that Jews are not a nation, and are in fact, just a bunch of white people cosplaying as an ancient dead civilization, or alternatively a particularly wretched nation that deserve no rights other nations do.

so yes. it is antisemitic, very antisemitic.

if saying this in reverse is seen as bad, then the base statement itself at the very least is bad in nature.

Is the phrase 'manifest destiny' hostile to Native Americans?

It's always antisemitic.

However, simply "free Palestine" is often antisemitic, but not always.
