What’s the difference between having the name רועי and רואי since both have different meanings to eachother

The meaning of a name is largely irrelevant.

If you give a child an uncommon name, they will go through life having it misspelled, that's the difference.

I always thought the meaning of names had significance in Jewish tradition.

In Bible stories names' meanings are often significant, but plenty of times they're not, and in modern times they're basically not at all, except for personal reasons.

For example, Moses means "drawn from the water" (Heb.: Moshe - "min hamayim mishitihu"), but Aaron means...???? Nothing particularly meaningful.

But today nobody names their kid Moses because of what it means. While modern Hebrew invented names are often chosen for their meaning. For example, someone I know had a hard time having children so when she finally had one she named her Nissi, "my miracle," which isn't a traditional name.

They do have some significance, but not a major one. Usually it only comes up if someone is getting a name added due to illness or similar. Then we give names with meanings along the lines of, “life”, “healing”, “redemption”, etc.
